it is a close up, so no need to stack, but focus not really on subject ( f.e. the eyes), that would be great, background colour dull, flash reflections should be avoided...try some different lightning, set focus point better, choose a higher aperture ( maybe 8)... and I'd get the ISO up to 200....my opinion
It would be great to know the exact reproduction rate, I guess it is 1:4...
